Reagent kit "BioPrime-HSV" is destined for definition of avidity index of specific G class immunoglobulin to HSV type 1 and 2 and for diagnostics of primary HSV infection. The method is based on detection of "early" low avidity IgG. Presence of low avidity antibodies indicates primary infection during the previous 2-3 months. The method is alternative to detection of virus-specific M class antibodies and does not require paired sera.
To define the avidity index if IgG antibodies each sample is tested in two parallel plate wells with immobilized herpes virus antigens. Specific to HSV human proteins antibodies interact with adsorbed antigens and produce immune complexes. After incubation with the tested serum experimental wells are additionally processed with denaturating solution, which destroys low avidity antibodies + antigen complexes, but does not influence high avidity complexes. Reduction of optical absorption in the well, processed with denaturating solution, in comparison with the control well, indicates presence of low avidity antibodies in the tested sample.
